Equipment available in the core includes:

2 Applied BioSystems 4000 Q TRAPs

 The Applied BioSystems 4000 QTRAP triple-quadrupole, linear ion trap hybrid MS provides maximum quantitative and qualitative performance with UPLC capability available from an in-line Shimadzu Prominence system.   A second set of Shimadzu pumps can be used for automated on line sample extraction using solid phase extraction.  The system has a TurboV ionization source with an electrospray or atmospheric pressure chemical ionization probe capable of positive and negative ionization mode, allowing detection of a wide range of molecules.  A photodiode array detector is also available prior to mass spectral analysis.  Sample analysis and instrument control will be conducted with Analyst software.  The combination of true triple-quadrupole and sensitive ion trap scan modes in a single LC/MS/MS run by the QTRAP allows for results that would otherwise require multiple analyses on multiple MS platforms.  The system also has LightSight™ Software that is excellent for finding drug metabolites.  The software automatically creates highly optimized methods that improve metabolite detection by constructing compound specific multiple reaction monitoring-information dependent acquisition methods (pMRM-IDA) where the survey scan will provide MS/MS data for potential metabolites.  The MS/MS spectra generated will be analyzed for structural elucidation by opening MS/MS spectra in ACD/MS Processor software directly from LightSight™.
 

Applied BioSystems 5500 Q TRAP

The Applied BioSystems 5500 QTRAP triple quadrupole linear ion trap hybrid mass spectrometerprovides the best quantitative and qualitative performance with UPLC capability available from an in-line Shimadzu Prominence system. It performs all the scan modes of the 4000 Q-TRAP and has a higher scan rate for more sensitive ion trap scan modes including MRM3 quantitation for analytes requiring greater sensitivity with high backgrounds.  The system has a TurbroV ionization source with both electrospray and atmospheric pressure chemical ionization probes capable of positive and negative ionization mode.  Sample analysis and instrument control will be conducted with Analyst software.  The system also has LightSight™ Software that is excellent for finding drug metabolites.  The software automatically creates highly optimized methods that improve metabolite detection by constructing compound specific multiple reaction monitoring-information dependent acquisition methods (pMRM-IDA) where the survey scan will provide MS/MS data for potential metabolites.  The MS/MS spectra generated will be analyzed for structural elucidation by opening MS/MS spectra in ACD/MS Processor software directly from LightSight™.
 

ThermoElectron Trace DSQ GC/Mass

This is a highly sensitive automated GC/MS instrument with an autosampler and split/spiltless injector and used for molecules requiring a gas chromatographic interface for separation. It can be used for electron impact or chemical ionization in positive and negative modes.  Instrument control and data analysis is afforded by Xcalibur software.
 

Agilent HPLC system

For molecules that can be quantified by HPLC the Core has two different HPLC systems. One is an Agilent 1100 quaternary gradient system with autosampler and a photodiode array and fluorescence detectors controlled with Chemstation software.
 

Computing and peripheral equipment 

The facility has two computers for data analysis.  Data from the instrumentation is transferred to the workstation computers where it can be manipulated by the staff or individual investigators.  Data is transferred for storage on servers that are maintained by the Advanced Computing Center at OHSU.  The facility has a Savant Speedvac system for sample preparation, a refrigerator and freezers for sample storage and an ultralow freezer for sample storage.
