We have a variety of shared synthesis, purification, and analytical instrumentation that can be used on a fee-for-use basis. Some Instrumentation requires prior user training by Core Staff before it can be reserved on iLab. User training can also be scheduled on iLab.

Automated Peptide Synthesizer - Liberty Blue

Automated Microwave Peptide Synthesizer

Requires user training

Our Automated Microwave Peptide Synthesizer is a CEM Liberty Blue,  located in LBRB 553J.
Features include

 

  • Peptide synthesis from 0.005 - 5 mmol scale.
  • Cycle times as low as 4 min (0.1 mmol scale).
  • Reaction vessels suitable for standard and high-swelling resins of 100-250 mesh.
  • Modifiable cycles for use with most peptide coupling reagents and deprotection solutions (optimized for DIC/Oxyma/Piperidine).
  • Microwave-assisted SPPS, up to 90 °C.

Included in the usage are standard amino acids, dimethylformamide, DIC, Oxyma, and piperidine. Necessary consumables are available in the Core for their pass-through costs. Resins are the user's responsibility.

User Training Fee

User training is carried out by core staff and includes 24-hour usage of the Automated Microwave Peptide Synthesizer. User training usually takes 90 minutes.

Consumables

The CEM Liberty Blue requires a special type of conical tube for the amino acid solutions. The use of these tubes is mandatory.

Fritted syringes and caps can be used for storage and further processing of the resin after synthesis. Their use is optional.

Microwave Reactor

Requires user training

Our Microwave Reactor is a Biotage Initiator+, located in LBRB 553J.
Features Include:

  • Heating reactions to high temperatures and pressures of up to 300 °C and 30 bar, allowing faster reaction times and higher concentrated reaction mixtures.
  • Automated sample processor for programming and performing sequential reactions in quick succession.
  • Four different vial sizes, 0.2 mL to 20 mL.
  • Easy-to-use large touchscreen display

Necessary consumables are available in the Core for their pass-through costs.

Our Parr Hydrogenator is located in LBRB 570.
Features include

  • Hydrogenation under hydrogen pressures up to 50 PSI.
  • Hydrogenation of volumes from 5 mL to 250 mL.

There are no necessary consumables for the Parr Hydrogenator.

User training is carried out by core staff and includes a 24-hour usage of the Parr Hydrogenator. User training usually takes 15 minutes.

Features include:

  • Automated normal and reverse phase column chromatography.
  • Various prepacked normal-phase column sizes, 5 g to 340 g.
  • Various derivatized and reverse-phase column sizes for special compounds.
  • Purification of milligrams to grams of material.
  • Diode array UV detector that can highlight multiple wavelengths.
  • Automated liquid gradient handling, including the option for mobile phase modifiers such as acids or bases.

Prepacked normal-phase columns are available in the Core for their pass-through costs. Reverse phase or derivatized columns must be ordered directly from Biotage.

Our lyophilizer is a Labconco FreeZone 2.5 L, located in LBRB 553J.
Features include

  • Refrigerated collector cooled to -84 °C, suitable for samples with low eutectic points including acetonitrile.

There are no necessary consumables for the lyophilizer.
Please inform Core Staff before lyophilizing samples containing solvents other than acetonitrile and water.

There are no fees associated with the use of the lyophilizer.

Refrigerated Centrifugal Concentrator (Speed Vac)

Does not require user training, only a short introduction at first use.

Our Speed Vac is a Labconco Refrigerated CentriVap Vacuum Concentrator, located in LBRB 572.
Features include

 

  • Rapid concentration of multiple samples, using centrifugal force, vacuum, and controlled temperature.
  • Cooling to -4 °C.
  • Heating to 100 °C
  • Rotors for 1.5 ml Eppendorf tubes and 15 ml conical tubes.

 

There are no necessary consumables for the Labconco Refrigerated CentriVap Vacuum Concentrator.
Please inform Core Staff before concentrating samples containing solvents other than acetonitrile and water or samples with more than 0.1 % acid.

Nuclear Magnetic Resonance (NMR) Core Facility 

Requires user training

Our NMR core facility has a Bruker 400 MHz Avance NEO NanoBay spectrometer, a powerful analytical tool routinely used for structural biology research. Housed in the Department of Chemical Physiology & Biochemistry, room number LBRB 670, the spectrometer is equipped with a broadband iProbe.

Features include

  • 24 - position Sample Case automation system.
  • Broad band iProbe.
  • Standard 1H/13C experiments (1D/2D) for monitoring reaction progress, checking purity, and determining molecular structure and concentration.
  • Advanced 2D NMR experiments (COSY, HSQC, HMBC, DEPT, NOESY/ROESY) for further structural elucidation.
  • Multi-nuclear NMR analysis (31P, 19F, 15N and other)

Wilmad Precision NMR tubes model numbers 527-PP-7 (7”) and 527-PP-8 (8”) are recommended as lower quality tubes not only result in poor spectra by introducing large spinning bands and bad line shape but also damage the probe.
