GI Stromal Tumor (GIST)
Test Description
KIT and PDGFRA gene exons are amplified by PCR and screened for mutations using denaturing HPLC (Transgenomic WAVE™ system), or high resolution melting curve analysis. Detected mutations are confirmed by standard bi-directional DNA sequencing. Testing is performed using a serial approach depending on primary tumor location.
Initial testing
Gastric tumor: KIT Exon 11
All other sites: KIT Exons 9 & 11
Additional testing performed if initial testing is negative
KIT exons (9), 13 & 17
PDGFRA exons 12, 14 & 18
